Abstract:
As a result of laboratory studies, the efficiency of applying standard and modified clonal micropropagation protocols for three selected poplar genotypes was assessed. Conditions were optimized at all stages: sterilization, culture initiation (MS medium), multiplication (WPM medium with 0.3 mg/L 6-BAP), rooting (½ MS with IBA), and ex vitro adaptation in a peat:vermiculite substrate (3:1). The influence of genotype on sterilization efficiency was evaluated: explant contamination rates varied from 30 % to 60 % among different cultivars. The highest multiplication coefficient (5.2) and the lowest shoot height (12.8 mm) were found in the hybrid P. alba × P. tremula, while P. 'Bashkirsky' exhibited the maximum shoot height (20.8 mm) and the lowest multiplication rate (2.8). The rooting frequency of regenerants across different poplar genotypes was relatively high, ranging from 70 % to 95 %.
